The Leica TCS SP2 is a point scanning confocal microscope, based around an inverted DMIRE stand. The system has three photomultipliers for fluorescence imaging with an additional one for transmitted light. It can allow users a wide choice of fluorochrome dyes including: DAPI, FITC, TRITC, TEXAS RED and CY5. Live-cell experimentation on this system is possible through the use of a stage-top live-cell incubation chamber where both temperature and CO2 concentration can be set and monitored.
A unique feature of the system is the ability to spectrally scan a specimen’s emission wavelength at a given laser excitation line, and thus determine whether the signal originates from the fluorochrome used or from background autofluorescence or bleethrough.
The operating software offers FRAP and FRET with built in ‘Wizards’ to help new users.
